Voor deze test wordt met een wattenstaafje een uitstrijk genomen uit de neus en keel. Bij de PCR-test een deel van het genetisch materiaal binnen in het virussterk vermenigvuldigd, om het aan te kunnen tonen. De PCR-test heeft een hoge testgevoeligheid.

Dat betekent dat de PCR-test het virus ook oppikt bij iemand die nog weinig virus heeft of weinig virus meer bij zich draagt. Om de test zo gevoelig mogelijk te maken wordt eerst een specifiek stukje van het genetisch materiaal vermeerderd gekopieerd met de zogenoemde polymerase-kettingreactie, voor de meeste mensen inmiddels bekend als PCR. De test kan dus geen positief resultaat geven bij andere virussen, zoals bijvoorbeeld een griepvirus of andere verkoudheidsvirussen. Tijdens elke cyclus wordt het genetisch materiaal verdubbeld, totdat het PCR-apparaat het detecteert.

Dus hoe meer virus aanwezig is, hoe minder verdubbelingen nodig zijn om die drempelwaarde te halen.
